Abstract

Se propone mejorar el diagnóstico molecular de las bacterias del orden Rickettsiales (familias Anaplasmataceae y Rickettsiaceae), que son transmitidas por garrapatas. Materiales y Métodos: se desarrolló y validó una membrana de hibridización reversa en línea (RLBH) para la identificación conjunta de bacterias de los géneros Ehrlichia, Anaplasma y Rickettsia. Se analizaron garrapatas recolectadas del ambiente (dragging), de perro, de humanos, de carpinchos provenientes de las provincias de Chaco, Corrientes y Santiago del Estero, Argentina. Resultados: la membrana tiene un buen poder discriminador para Anaplasma y Ehrlichia, para las Rickettsias en cambio, la discriminación no es completa. Se verificó la presencia de candidatus R. sp. argentina en Amblyoma parvum y se detectó R. bellii en una muestra de A. dubitatum. Conclusiones: La herramienta desarrollada resulta útil como metodología diagnóstica para evaluar la emergencia de las enfermedades bacterianas transmitidas por garrapatas.

We propose to improve the molecular diagnosis of bacteria from the order Rickettsiales (Anaplasmataceae and Rickettsiaceae families), which are tick-borne. Materials and Methods: a reverse line bloot hibridization membrane (RLBH) was developed and validated for the simultaneous identification of the genera Ehrlichia, Anaplasma and Rickettsia. Ticks collected from dogs, humans, capybaras and in host seeking (dragging) from Chaco Corrientes and Santiago del Estero (Argentina) were analized. Results: the membrane resulted in a high discrimination power for Anaplasma and Ehrlichia. For the genus Rickettsias instead discrimination was not complete. We verified the presence of candidatus R.sp. argentina in Amblyoma parvum and we detected R. bellii in A. dubitatum. Conclusions: The developed tool resulted useful for diagnosis and the study of the emergence of tick-borne bacteria.
