Actividad inmunomoduladora de péptidos de Amaranthus hypochondriacus

Abstract

Los objetivos del trabajo fueron obtener fracciones peptídicas a partir de la semilla de A. hypochondriacus y evaluar su efecto inmunomodulador haciendo uso de cultivo de células intestinales Caco-2 ccl20:luc. Además se identificaron y sintetizaron los péptidos responsables de la modulación. Se evaluó el efecto inmunomodulador de un aislado proteico de A. hypochondriacus y de dos hidrolizados del mismo por acción de alcalasa con 23% y 30% grado de hidrólisis (GH), a diferentes concentraciones. Se utilizaron células epiteliales de colon humano transfectadas con el gen reportero luciferasa bajo el control del promotor ccl20 (Caco-2 ccl20:luc) altamente inducible por el estímulo pro-inflamatorio del Toll-Like Receptor 5 (cuyo agonista es flagelina (FliC)), siendo un indicador sensible de la respuesta inmune innata. Las células Caco-2 ccl20:luc fueron incubadas con aislado e hidrolizados a diferentes concentraciones, y estimuladas en forma conjunta con FliC por 4 h. Los resultados mostraron una actividad inhibitoria que aumentó con la concentración (comportamiento dosis-respuesta) y con el GH.
