Compatibilidad de Lactobacilos benéficos vaginales (LBV) con ingredientes farmacéuticos activos

Abstract

Los lactobacilos son los microorganismos predominantes en el microbioma vaginal de mujeres sanas, y responsables de mantener el equilibrio ecológico y prevenir el ingreso y sobrecrecimiento de oportunistas y patógenos que producen infecciones. LBV pueden incluirse en el diseño de fórmulas para la prevención y/o tratamiento de infecciones urogenitales combinados con otros principios activos de aplicación vaginal, que podrían favorecer o complementar su efecto benéfico. La selección de estos componentes debe realizarse criteriosamente para potencializar la funcionalidad del producto final, y que sean compatibles para evitar efectos antagónicos. El objetivo de este trabajo fue evaluar la compatibilidad de 5 cepas de LBV (L. gasseri CRL1263, L. gasseri CRL1509, L. reuteri CRL1324, L. salivarius CRL1328 y L. rhamnosus CRL1332) previamente aislados de vagina de mujeres y seleccionados en nuestro grupo de investigación por sus propiedades benéficas, con diferentes principios activos (antimicrobianos, regeneradores de epitelio, vitaminas y hormonas). La compatibilidad de los microorganismos con principios activos se realizó aplicando el método de microdilución y/o en medio sólido, de acuerdo a ISO/DIS10932/IDF223 con modificaciones. Todas las cepas resultaron compatibles con las siguientes sustancias: 16 µg/ml de econazol, ketoconazol, miconazol, clotrimazol y nistatina (antimicóticos), 2500 µg/ml de tinidazol, 10000 µg/ml de alicina, 12000 µg/ml de vitamina A, 512 µg/ml de estriol y 15000 µg/ml de centella asiática. Asimismo mostraron resistencia a elevadas concentraciones de metronidazol. L. gasseri CRL1509, L. reuteri CRL1324, L. salivarius CRL1328 y L. rhamnosus CRL1332 fueron incompatibles con clindamicina. En el caso de neomicina, L. reuteri CRL1324 resultó incompatible, mientras L. salivarius CRL1328 fue resistente a 1 µg/ml, L. rhamnosus CRL 1332 a 4 µg/ml y L. gasseri CRL1263 y 1509 a 16 µg/ml. L. reuteri CRL 1324 y L. salivarius CRL 1328 resisten a las menores concentraciones de nitrofurazona (7,81 µg/ml), mientras L rhamnosus CRL1332 fue compatible con 125 µg/ml. Las cinco cepas de LBV se pueden combinar con vitamina C (7810 µg/ml) y ácido bórico (12500 µg/ml). Los resultados obtenidos permiten seleccionar los componentes activos compatibles con cepas de lactobacilos con propiedades benéficas para avanzar en el diseño de fórmulas de aplicación vaginal.
