Farmacogenomica: Camino hacia una medicina personalizada

Abstract

La variabilidad individual en la eficacia y seguridad de los medicamentos representa uno de los principales desafíos en la práctica clínica actual. Un fármaco que ha demostrado ser eficaz en muchos pacientes, a menudo, puede no resultar adecuado para algunos otros por no producir el efecto esperado. Además, en repetidas ocasiones una droga puede causar serias reacciones adversas (RAM), que incluso, en algunos casos, llegan a comprometer la vida del paciente. Si bien desde hace muchos años se conoce la existencia de esta amplia heterogeneidad en la respuesta a drogas, el origen de la misma resulta aún difícil de elucidar. Asimismo, la frecuencia y el grado de severidad de las reacciones adversas constituyen un factor decisivo en el proceso regulatorio para la aprobación de una droga como así también para su retiro del mercado una vez que la droga ya ha sido aprobada. Por este motivo, en los últimos años, el reconocimiento de los factores que causan el amplio espectro de respuesta a un tratamiento ha cobrado gran relevancia en la práctica clínica y en el desarrollo de nuevos agentes terapéuticos. En este sentido los estudios farmacogenéticos y farmacogenómicos conducen a identificar biomarcadores que contribuyen a optimizar la conducta terapéutica en el camino hacia una medicina personalizada.
