Los procesos LADME y la nanotecnología

Abstract

El desarrollo de medicamentos cuyos principios activos sean transportados de manera selectiva hacia sitios específicos del cuerpo (terapias dirigidas), de manera de maximizar la seguridad y eficacia del tratamiento, es uno de los grandes desafíos actuales de las Ciencias Farmacéuticas. Hoy día gran número de fármacos de potencial uso en terapéutica poseen características biofarmacéuticas inadecuadas, las cuáles dificultan su llegada a la biofase en tiempo y forma. Una estrategia para mejorar las limitaciones intrínsecas de tales activos es el empleo de vehículos o vectores que mejoren sus perfiles biofarmacéutico y farmacocinético, sorteando inconvenientes tales como: escasa solubilidad, inestabilidad fisicoquímica, rápida eliminación o incapacidad del fármaco de atravesar ciertas barreras biológicas. Adicionalmente, el empleo de vehículos adecuados permitiría limitar la toxicidad inespecífica (off target, fuera del blanco molecular) del tratamiento. Los objetivos anteriores no siempre se consiguen empleando los vehículos y formas farmacéuticas convencionales, planteándose la necesidad de recurrir a otros sistemas innovadores, que denominaremos sistemas de liberación avanzada o de última generación.
