Er fullstendig selektiv inhibering av CLK1 en mulighet?

Abstract

Cdc2-like kinase 1 (CLK1) er en proteinkinase som er involvert i pre-mRNA spleising. Den er sammenkoblet med flere sykdommer. Derfor er inhibering av CLK1 er et svært aktuelt tema, da dette kan forhindre flere lidelser. Ved å gjøre et litteratursøk blir det forsøkt å svare på problemstillingen: "Er selektiv inhibering av CLK1 mulig, og hvis det er mulig, hvilken inhibitor er best for CLK1?" Dette blir gjort ved å først se på CLK1 sin funksjon i cellen, og ulike inhibitorer til CLK1. I diskusjonen blir teorien brukt for å svare på problemstillingen. For å finne ut hvor selektiv inhibitorene er blir IC50-verdiene for CLK1 og DYRK1A sammenlignet. Videre blir det konkludert med at selektiv inhibering av CLK1 ikke er et enkelt mål, hvor forbindelse 5 er den som så langt har best dokumentert effekt. Selektiv inhibering er vanskelig å få til siden CLK1 deler egenskaper med andre enzymer, som DYRK1A.
