Abstract

Amyotrofische laterale sclerose (ALS), progressieve spinale spieratrofie en primaire laterale sclerose zijn ziekten in de motorische voorhoorncel. De aandoeningen worden vooral op klinische gronden van elkaar onderscheiden. Maaike van der Graaff bekeek of motorische en niet-motorische neuronen in de hersenen (zoals structuren betrokken bij geheugen of gedrag) zijn aangetast bij patiënten met een van de drie aandoeningen. Van der Graaff onderzocht dit met MR-spectroscopie en MR-diffusie tensor imaging, net nadat de diagnose was gesteld. Bij alle patiënten bleek al vroeg in het ziekteproces dat motorische en niet-motorische neuronen waren aangetast. Het klinische onderscheid tussen de drie ziektebeelden lijkt dus niet terecht. Van der Graaff volgde in de zes maanden daarna met dezelfde technieken de verdere aantasting van de hersenen. Het corpus callosum (hersenbalk die de twee hersenhelften met elkaar verbindt) lijkt hierbij een belangrijke rol te spelen.
