Abstract

Het cytoskelet bestaat uit eiwitketens (polymeren), waarvan de lengte fluctueren. Het functioneert onder andere als ‘wegennet’ en reguleert het transport van eiwitten naar specifieke plaatsen binnen de cel. Een aantal eiwitten is van belang voor celbeweging, waaronder Rac1. Micha Nethe beschrijft signalering van Rac1 en moleculaire aansturingsmechanismen die Rac1 gebruikt om celbeweging en hechting te beïnvloeden. Signalering is van belang voor migratie van witte bloedcellen naar de plek van een ontsteking. Endotheelcellen zorgen ervoor dat deze cellen via de bloedvatwand de plaats van de ontsteking kunnen bereiken. Na het verlenen van doorgang moet het contact tussen de endotheelcellen onmiddellijk worden hersteld, zodat er geen ‘lek’ ontstaat dat het achterliggende weefsel alsnog kan beschadigen. Verloopt dit niet goed, dan kunnen ontstekingsziektes ontstaan als astma, Crohn en reuma. Ook kan zo’n lek zorgen voor het uitzaaien van kankercellen. Nethe onderzocht nieuwe eiwitten die aan Rac1 binden.
