Abstract

Bij een infectie met hiv is het zogeheten envelop-eiwit verantwoordelijk voor het versmelten van het membraan dat het virus omhult met het membraan van de gastheercel. Vervolgens kan het erfelijk materiaal van het virus de cel binnendringen en overnemen. Het envelop-eiwit bestaat uit verschillende eiwitten die samen een soort scharnier vormen. Dirk Eggink beschrijft een nieuwe klasse hiv-remmers in de vorm van peptiden (kleine stukjes eiwit) die aan het scharnier binden en zo versmelting van membranen voorkomen. Helaas kan hiv ook hiertegen resistentie ontwikkelen. Eggink ging na welke mutaties in hiv verantwoordelijk zijn voor resistentie tegen fusieremmers T1249 en T2635 en welke mechanismen zorgen voor deze resistentie. De remmer T2635 werkt het best: voor resistentie zijn meerdere mutaties in het envelop-eiwit nodig. Bovendien bleek het virus, ook als eenmaal resistentie was ontstaan, minder goed in staat om nieuwe cellen te infecteren.
