Abstract

En esta Tesis se presenta un modelo (teórico-computacional) básico de la electroforesis capilar por zona (CZE) que involucra moléculas cargadas y pequeñas, el cual se valida con datos experimentales propios y de la bibliografía. Este modelo (no-ideal y lineal) provee predicciones cuantitativas de interés práctico para la interpretación del electroferograma de un analito o de una mezcla de analitos. Es posible ampliar las capacidades del modelo (generando los modelos extendidos: LLCEM y PLLCEM) para aplicarlo a analitos más complejos como péptidos y proteínas. Se incluye aquí el fenómeno de regulación de cargas, lo cual conduce a conclusiones relevantes sobre el cálculo de la carga neta, el radio hidrodinámico, la forma, la hidratación, el pH cercano a la molécula y la estimación de las variaciones en los pK de los grupos ionizantes. Se pone énfasis en el hecho que estos parámetros están físicamente acoplados. Asimismo, los modelos extendidos son útiles para estudiar el proceso de desnaturalización y la estabilidad conformacional de proteínas bajo la acción de diferentes agentes desnaturalizantes, a partir de medidas sencillas de la movilidad electroforética que se obtienen mediante CZE. Se demuestra que la CZE, además de ser una herramienta analítica ampliamente usada, tiene un alto potencial para la estimación de las propiedades fisicoquímicas como radio hidrodinámico, coeficiente de difusión, carga eléctrica efectiva, potencial zeta y forma e hidratación de los analitos, cuando el electroferograma se modela e interpreta a través de las ecuaciones de los fenómenos de transporte acopladas a los efectos electrocinéticos.

This Thesis presents a theoretical-computational model of capillary zone electrophoresis (CZE) involving small charged molecules, which is validated with experimental data. This model (non-ideal and linear) provides quantitative predictions of practical interest for electropherogram interpretations involving an analyte or a mixture of analytes. It is possible to extend this basic model (yielding the so-called extended models: LLCEM and PLLCEM) to more complex analytes like peptides and globular proteins, by including the charge regulation phenomenon, which leads one to relevant conclusions on the calculations of net charge, hydrodynamic radius, shape, hydration, pH near molecule and the estimation of pK variation of ionizing groups. Special emphasis is placed on the fact that these parameters are physically coupled. Also, the extended models may be useful to study the denaturizing process and conformational stability of globular proteins under the effect of different denaturant agents, from the measurement of simple electrophoretic mobilities as obtained from CZE. This Thesis shows that CZE, apart from being an analytical tool widely used, has a high power for the estimation of physicochemical properties like, for instance, hydrodynamic radius, diffusion coefficient, effective electrical charge, zeta potential and shape and hydration of analytes, when the electropherogram is modeled through the balance equations of transport phenomena coupled to electrokinetic effects.
