Optimización de la Producción de Eritropoyetina Humana Recombinante (rhEPO) Mediante el Cultivo de Células Animales

Abstract

En este trabajo se presentan los resultados obtenidos durante la optimización de la producción de eritropoyetina humana recombinante (rhEPO) en cultivo de células CHO (Chinese Hamster Ovary cells), partiendo de la selección de un nuevo clon productor, el estudio del efecto de aditivos y variables fisicoquímicas sobre el crecimiento, productividad y patrón de isoformas producidas, y finalmente, el diseño racional de medios de cultivos que permiten obtener una molécula de alta calidad glicosídica. Inicialmente se clonaron líneas celulares recombinantes productoras de rhEPO. Los clones seleccionados fueron adaptados a crecer en suspensión, en medios de cultivo de bajo contenido de suero y libre de proteínas, estudiándose el efecto que ejercen sobre el metabolismo diferentes metodologías de adaptación a suspensión, concentraciones de suero y sistemas de cultivo. Utilizando un diseño factorial se evaluó la influencia de la adición de aminoazúcares, la acumulación del ion amonio en sobrenadante y el efecto de la osmolalidad, sobre el crecimiento, la productividad y el patrón de glicosilación. Posteriormente, y mediante un diseño experimental de mezclas cruzadas con limitaciones, acoplado a un análisis multivariado de las respuestas (a través de las metodologías de superficies de respuesta o de redes neuronales artificiales), seguido de un proceso de optimización, se diseñaron dos medios de cultivo con el objetivo de optimizar la producción de rhEPO. Estos medios fueron utilizados durante el cultivo del clon productor en un biorreactor tipo tanque agitado, en modo continuo con perfusión, obteniéndose una molécula de rhEPO de alta calidad glicosídica.

In this thesis report, we present the results obtained during the optimization of the production of recombinant human erythropoietin (rhEPO) in Chinese Hamster Ovary (CHO) cell culture, involving the selection of a new clone, the study of the effect of new additives and physicochemical variables on growth, productivity and isoform pattern, and finally, the rational design of culture media in which highly glycosylated rhEPO molecules are obtained. Firstly, recombinant cell lines producing rhEPO were cloned. The selected clones were adapted to grow in suspension mode in both low-serum-content medium and protein-free medium. The effect of different methodologies to adapt cells to suspension growth, as well as different serum concentrations and culture systems were studied. The effect of addition of aminosugars, as well as the accumulation of ammonium ion in the supernatant were evaluated using a factorial design. The variability in supernatant osmolality was also analyzed. Two culture media which improve rhEPO production were designed using a constrained crossed mixture experimental design, followed by multivariate analysis of responses: firstly, through a response surface methodology, and later, by artificial neural networks, coupled to an optimization process. The formulated media were used to cultivate the selected clone in a stirred tank bioreactor, in a perfused continuous mode, accomplishing the pursued goal, a highly glycosylated molecule.
