Estandarización de un protocolo para el aislamiento, cultivo y mantenimiento de células primarias de queratinocitos in vitro

Abstract

El aislamiento y cultivo de queratinocitos epidérmicos en ratón o ratas es una técnica común que permite fácilmente la manipulación genética. Sin embargo, debido a su limitada esperanza de vida en el cultivo, los experimentos que utilizan queratinocitos primarios requieren un gran número de animales, tiempo y dinero. Es por esto que este proyecto de investigación buscó estandarizar un protocolo para el aislar la mayor cantidad de queratinocitos in vitro, y que permitieran mantenerse por más tiempo este tipo de líneas celulares en condiciones in vitro. Para esto, se usó ratas Wistar en estadio P0 a P2, ya que en estos estadios se encuentran mayor cantidad de queratinocitos basales no diferenciados. Se realizó un tratamiento enzimático con tripsina durante 18h a 4°C para facilitar el aislamiento de los queratinocitos
