Caracterización cinética de la betalactamasa de pseudomonas fluorescens producida de forma recombinante

Abstract

Debido a la importancia de las betalactamasas en el desarrollo de mecanismos antimicrobianos, el objetivo de este proyecto es realizar una caracterización cinética de la betalactamasa recombinante de Pseudomonas fluorescens mediante su producción en el organismo E. Coli BL21(DE3), el cual se llevó a cabo actualmente en la Universidad Icesi. Para esto, se transformaron bacterias E. Coli BL21 (DE3), con el plásmido pET21b- Pfluor-Betalac el cual contiene el gen de interés que codifica por la betalactamasa. Posteriormente, se extrajó y se purificó la betalactamasa recombinante por medio de cromatografía de afinidad con resina de níquel, y por último se realizó ensayos cualitativos y cuantitativos de actividad enzimática con rojo de fenol, además se deseó determinar los parámetros cinéticos de reacción enzimática de la betalactamasa aislada, como la constante de Michaelis (KM) y la velocidad máxima de catálisis (Vmax), empleando ampicilina como sustrato.
