El trastorno depresivo mayor es una enfermedad compleja en la cual interactúan factores psicosociales, genéticos y biológicos. La metaloproteinasa de matriz 9 (MMP-9) es una colagenasa que participa en la degradación de la matriz extracelular, la memoria y el aprendizaje. En este trabajo se estudió la expresión de MMP-9 en el hipocampo de un modelo murino de obesidad e inflamación, así como en la corteza prefrontal de pacientes con depresión mayor. Los ratones con dieta rica en grasa y/o lipopolisacárido mostraron un comportamiento de tipo ansioso-depresivo, y un incremento en los niveles de MMP-9 en el grupo de dieta rica en grasa. En las muestras humanas postmortem se observó un aumento en la expresión de MMP-9 en las mujeres de las cohortes 'no tratadas' y 'suicidio'. En conclusión, en el modelo murino no parece existir una correlación entre los niveles hipocampales de MMP-9 y el fenotipo ansioso-depresivo. En las muestras humanas, el incremento en la expresión cortical de MMP-9 en las mujeres de los grupos 'no tratadas' y 'suicidio', apoyaría la relación entre MMP-9 y severidad de la depresión. Los resultados en muestras humanas apoyarían la relevancia de la MMP-9 como potencial diana terapéutica, y/o biomarcador de la enfermedad.
Major depressive disorder is a complex illness in which psychosocial, genetic, and biological factors interact. Matrix metalloproteinase 9 (MMP-9) is a collagenase that is involved in the degradation of the extracellular matrix, as well as in memory and learning. This study examined the expression of MMP-9 in the hippocampus of a male murine model combining obesity and inflammation, and in cortical samples from patients diagnosed with major depression. Mice displayed anxious-depressive behavior in behavioral tests, and increased MMP-9 levels in the high-fat diet group. However, the human samples showed an increase in MMP-9 expression in depressed women in the 'untreated' and 'suicide death' cohorts compared to their respective control groups. In conclusion, in the murine model, there does not appear to be a relevant correlation between hippocampal MMP-9 levels and the anxious-depressive phenotype. In the human samples, the increased cortical MMP-9 expression in the groups of women 'untreated' and 'suicide death' might support the association between MMP-9 and the severity of depression. The results obtained in human samples suggest that MMP-9 could be a potential a therapeutic target and/or a biomarker of this pathology.
