Abstract

RESUMEN : La senescencia celular, un proceso implicado en el mantenimiento de la integridad del organismo ante factores perturbadores de la homeostasis celular, tiene un papel fundamental durante el desarrollo. Sin embargo, cada vez hay más evidencia de su implicación en la aparición y agravamiento de enfermedades neurodegenerativas, como la enfermedad de Alzheimer (EA) y la enfermedad de Parkinson (EP). Son múltiples los mecanismos celulares que intervienen en la aparición de la senescencia, entre los que destacan los cambios en la expresión génica, la disfunción mitocondrial y el estrés oxidativo, las alteraciones de la autofagia, de la actividad lisosómica y de la proteostasis, así como los cambios epigenéticos. Además, la alteración en el plegamiento de proteínas, como es el caso la proteína tau fosforilada, las placas de bamiloide (Aβ) y la α-sinucleína, que caracterizan a nivel anatomopatológico numerosas enfermedades neurodegenerativas, también está implicada en el desarrollo de la senescencia celular. Se trata de un tema cuya comprensión e investigación se encuentran en plena expansión y en este trabajo se resume la evidencia científica disponible sobre la implicación de la senescencia celular en la patogenia y progresión de algunas patologías neurodegenerativas.

ABSTRACT : Cellular senescence, a process implicated in the maintenance of the integrity of the organism in the presence of factors that can alter cellular homeostasis, has a crucial role during development. However, there is increasing evidence of its implication in the onset and aggravation of neurodegenerative diseases, such as Alzheimer's disease (AD) and Parkison's disease (PD). Among the multiple cellular mechanisms implicated in the onset of senescence are changes in gene expression, mitochondrial dysfunction and oxidative stress, alteration of autophagy, lysosomal activity and proteostasis, as well as the implication of epigenetic changes. Also, alterations in the folding of proteins such as phosphorylated tau protein, b-amyloid (Aβ) plaques and α-synuclein, that characterize several neurodegenerative diseases at an anatomopathological level, have been implicated in the development of cellular senescence. It is a theme whose understanding and research are in full expansion and this work aims to summarize the scientific evidence that is available.
