Abstract

RESUMEN : La melatonina es la principal hormona sintetizada y secretada por la glándula pineal, en función del ciclo luz/oscuridad, dando lugar ello a una de sus funciones más importantes, la regulación de los ritmos circadianos. Sin embargo, otra de las acciones importantes descritas de la melatonina es sus efectos oncostáticos, inhibiendo la génesis y desarrollo de una gran variedad de tumores, particularmente hormonodependientes. El presente trabajo describe y efectúa una revisión sistemática de ensayos clínicos en los que se utiliza melatonina, observándose algunos de los principales efectos antitumorales ejercidos por esta hormona, que incluye: acciones antiestrogénicas; modulación del ciclo celular; efectos antioxidantes; antiangiogénesis y efectos inmunoestimuladores. En la gran mayoría de los ensayos clínicos, la melatonina presenta efectos beneficiosos sobre los pacientes con cáncer avanzado, como aumento de su supervivencia, aumento de la tasa de regresión tumoral, control sobre el avance de la enfermedad y disminución de los efectos adversos provocados por otros tratamientos como la radioterapia o quimioterapia.

ABSTRACT : Melatonin is the main hormone synthesized and secreted by the pineal gland, depending on the light/dark cycle, leading to one of the most important actions carried out by melatonin, the circadian rhythms´ regulation. However, another of the important actions described by melatonin is its oncostatic effects, inhibiting the genesis and development of a wide variety of tumors, particularly hormone-dependent. The present work describes and carries out a systematic review of clinical trials in which melatonin is used, observing some of the main antitumor effects exerted by this hormone, which includes: antiestrogenics actions; cell cycle modulation; antioxidants effects; antiangiogenesis and immunostimulatory effects. In the vast majority of clinical trials, melatonin achieves beneficial effects on patients with advanced cancer, such as increased survival time, increased the tumor regression rate, control over the progression of the disease and decreased the adverse effects caused by other treatments such as radiation therapy or chemotherapy.
