Abstract

RESUMEN: La reducción genómica está basada en la suposición de que sólo una parte de los genes bacterianos son necesarios para sobrevivir en un ambiente constante, como el que se proporciona en un medio de cultivo de laboratorio. Una célula con una dotación genética mínima, pero óptima para crecer en un determinado ambiente, debería tener ventajas frente a su predecesora (célula de partida), como por ejemplo, mayor estabilidad genética debido a la eliminación de ADN parásito (fagos, transposones y genes de función desconocida) y mayor producción de biomasa o de ciertos compuestos de interés. Pero sobre todo, debería ser un genoma más fácil de controlar, lo cual es muy interesante desde el punto de vista industrial y experimental. En el presente trabajo se expone el diseño y puesta a punto de diversas técnicas que permiten la manipulación genética para Synechococcus elongatus (Se7942), como por ejemplo: la puesta a punto de un protocolo de eliminación genética eficaz en Se7942, el establecimiento de un protocolo optimizado de conjugación interphyla a cianobacterias (desde E.coli hasta Se7942), el diseño y obtención de un juego de vectores lanzadera para Se7942, así como el estudio del proceso de transformación natural en Se7942. Gracias a las técnicas desarrolladas y puestas a punto en este trabajo se ha logrado obtener un primer chasis autotrófico reducido de Se7942 (1,2% de reducción genómica), el cual muestra mejores eficiencias de conjugación y transformación natural al ser utilizado como receptor en dichos procesos.
