Evaluación de la biocompatibilidad in vitro de materiales para uso biomédico

Abstract

[ES] El incremento de la expectativa de vida de la población conlleva un aumento en la demanda de terapias encaminadas a corregir el deterioro de órganos y tejidos; sin embargo, las soluciones hoy en día siguen estando limitadas al trasplante de estos. La escasez de donantes y el riesgo de padecer rechazo inmunológico frente al trasplante han obligado a desarrollar alternativas terapéuticas basadas en la ingeniería tisular. Un “constructo” de ingeniería tisular incluye células, biomateriales y señales químicas o físicas reguladoras de la función celular. Por esta razón, actualmente existe una gran demanda de biomateriales para estas aplicaciones, siendo el principal requisito que deben cumplir, su carácter biocompatible. En este trabajo, utilizando una línea celular de fibroblastos de ratón, se ha estudiado la biocompatibilidad in vitro de cinco nuevos derivados del poliuretano, mediante la evaluación de su citotoxicidad y su capacidad para constituir un sustrato de adhesión celular. Para ello, por una parte, se cuantificó la proliferación celular de cultivos realizados en medio extractivo obtenido de cada uno de los materiales y, por otra, se realizaron cultivos directamente sobre muestras de los materiales y se evaluó la adhesión y proliferación celular mediante microscopía electrónica de barrido. Dos de los cinco materiales han demostrado una buena biocompatibilidad in vitro al no mostrar indicios de citotoxicidad para la línea celular utilizada, además de permitir la adhesión, migración y proliferación de las células sobre el material con gran eficacia. Este estudio abre camino a la futura evaluación de la biocompatibilidad in vivo de estos materiales y sienta las bases para su futura aplicación en ingeniería tisular y biomedicina.
