Incertidumbre en métodos analíticos de rutina.

Abstract

According to the ISO Norm 17025, the accredited analytical laboratories should obtaintheir results accompanied by two basic quality parameters: traceability and uncertainty.Therefore, nowadays the analysts are increasingly urged to assess the traceability of theirresults and to estimate the uncertainty of these results. In this thesis, we have proposeddifferent methodologies to calculate uncertainty in analytical methods that are usedroutinely in the laboratory. These methodologies calculate uncertainty as a whole (i.e. bygrouping terms of uncertainty whenever possible) and use the information generated duringthe validation process and, specially, the information obtained in the assessment oftraceability. Moreover, the uncertainty can also be calculated using the informationgenerated during precision studies, during robustness studies and during the internal qualitycontrol.The advantage of the proposed methodologies is that they require few additional work tocalculate uncertainty because the analytical methods should always be validated beforeapplying them to the analysis of routine samples. We have developed the calculation ofuncertainty at one concentration level when traceability is assessed with several types ofreferences such as certified reference materials (CRM), reference methods andinterlaboratory studies. This uncertainty is calculated using the information obtained in theassessment of traceability. Moreover, this uncertainty can be calculated by using theinformation generated from control charts, precision studies and robustness studies. Thecalculation of uncertainty at one concentration level should only be applied in a restrictedconcentration range because it is assumed that both the method bias and precision are thesame in the whole concentration range. Para que los laboratorios de análisis puedan acreditarse según la norma ISO 17025, es necesario que los resultados analíticos vayan acompañados de dos parámetros de calidadbásicos: su trazabilidad y su incertidumbre. Esto ha hecho que, hoy en día, la verificaciónde la trazabilidad y el cálculo de la incertidumbre de los resultados analíticos sea cada vezmás importante. En esta tesis doctoral hemos propuesto diversas metodologías para calcularla incertidumbre en métodos analíticos que se utilizan de forma rutinaria en el laboratorio.Estas metodologías están basadas en calcular la incertidumbre globalmente (es decir,agrupando términos de incertidumbre siempre que sea posible) y en utilizar la informaciónobtenida durante el proceso de validación del método y, especialmente, durante laverificación de la trazabilidad.La ventaja de estas metodologías es que pueden aplicarse a todos los métodos de rutina yque requieren poco trabajo adicional ya que los métodos analíticos siempre deben validarseantes de aplicarlos al análisis de muestras de rutina. En concreto, hemos propuestoexpresiones para calcular la incertidumbre a un nivel de concentración cuando latrazabilidad de los resultados se verifica utilizando diversas referencias como los materialesde referencia o los ejercicios interlaboratorio. Para ello, se utiliza fundamentalmente lainformación obtenida al verificar la trazabilidad. Además, también puede utilizarse lainformación obtenida a partir de los gráficos de control, los estudios de precisión y losestudios de robustez. El cálculo de la incertidumbre a un nivel de concentración esaplicable en métodos de rutina con un intervalo restringido de concentraciones ya que seasume que el sesgo y la precisión del método es el mismo en todo el intervalo deconcentraciones.
