Abstract

En el Departament de Bioquímica de la Facultat de Medicina s'està portant a terme, des de fa un temps, una línia d'investigació basada en l'estudi del metabolisme del 2,3-bisfosfoglicerat, el cofactor de la fosfoglicerat mutasa. Recentment s´ha desenvolupat en el laboratori una altra línia de treball sobre el metabolisme de la glucosa 1,6-bisfosfat, el cofactor de la fosfoglucomutasa, basant-se en les analogies amb el compost esmentat anteriorment. La glucosa 1,6-bisfosfat, a més de cofactor de la fosfoglucomutasa, és un potent efector de diversos enzims del metabolisme de carbohidrats. L'estudi s'ha dedicat essencialment a les vies de síntesi i degradació del metabòlit, com també al seu paper com a regulador del metabolisme muscular. S'ha de remarcar que el treball que es presenta en aquesta Memòria ha estat l'inici d'aquesta línia d'investigació, ja que no hi havia cap antecedent en el laboratori. Amb aquests antecedents, el present treball es va dirigir cap a dos punts d'interès: En primer lloc, es va estudiar el paper de la glucosa 1,6-bisfosfat bibliografia, en algunes situacions no descrites a la bibliografia, concretament la contracció muscular i el dejuni. La contracció del múscul es un procés que transcorre amb una forta activació del flux glicolitic i encara no estan clars els factors que afecten de manera essencial l'activitat de la fosfofructoquinasa. Un altre estudi realitzat ha estat la determinació de les concentracions de glucosa 1,6-bisfosfat, fructosa 2,6- bisfosfat i activitats enzimàtiques relacionades en músculs amb diferent composició de fibres. El metabolisme d'aquests és força divers i la tendència actual és diferenciar cada cop més aquests tipus de fibres que constitueixen, en si mateixes, unes unitats bàsiques diferenciades. En segon lloc, s'ha iniciat l'estudi de les activitats enzimàtiques responsables de la degradació de la glucosa 1,6-bisfosfat, aspecte molt poc descrit a la bibliografia. S'ha considerat també el possible paper de la fosfoglucomutasa en aquest procés, a causa de la seva activitat glucosa 1,6-bisfosfat intrínseca. L'estudi es va iniciar en teixits de porc i es va continuar en teixits de rata, per tal de disposar d'un animal adequat per estudis "in vivo".
