Abstract

El clorpirifos (CPF) es un insecticida organofosforados ampliamente utilizado. Sus efectos neurotóxicos derivan principalmente de la inhibición de colinesterasas. Diferentes investigaciones han indicado efectos conductuales y cognitivos por exposición a CPF. En el presente trabajo, hemos estudiado los efectos neuroconductuales derivados de la exposición a una dosis aguda y dosis repetidas de CPF, en un modelo de ratón transgénico (Tg2576) portador de la mutación Swedish, responsable de una forma familiar de la enfermedad de Alzheimer. Se evaluaron los efectos sobre la actividad exploratoria, el aprendizaje y la memoria espacial y la coordinación motora. Se determinaron la actividad acetilcolinesterasa, los niveles de beta-amiloide (1-40, 1-42), la proliferación y diferenciación celular y la expresión de BACE1. Los resultados mostraron una mejora cognitiva en los Tg expuestos al CPF. Sin embargo, observamos un incremento en los niveles de A en los Tg tratados, que supondría a largo plazo un empeoramiento del proceso neurodegenerativo.

Chlorpyrifos (CPF) is a widely used organophosphate insecticide. Neurotoxic effects derive primarily from the inhibition of cholinesterase. Different studies have shown behavioral and cognitive effects of exposure to CPF. In the present study, we evaluated the neurobehavioral effects resulting from exposure to an acute dose and repeated doses of CPF, in a transgenic mouse model (Tg2576) carrying the Swedish mutation responsible for a familial form of Alzheimer's disease. We evaluated the effects on exploratory activity, learning and spatial memory and motor coordination. Acetylcholinesterase activity, levels of beta-amyloid (1-40, 1-42), proliferation and cell differentiation and expression of BACE1, were determined. The results showed cognitive improvement in Tg exposed to CPF. However, we observed an increase in the levels of A in the treated Tg, which would be a long term worsening of the neurodegenerative process.
