Avaliação da genotoxicidade do cádmio em duas espécies de Thlaspi

Abstract

Cadmium (Cd) is a non essential element and is a widespread environmental pollutant. Exposure to Cd can result in cytotoxic, carcinogenic and mutagenic effects. Mutagenesis can be assayed with the use of molecular techniques such as microsatellites and, measuring genetic instability, may be a powerful complement in toxicological evaluations. In this study the mutagenic/genotoxic effects of cadmium will be evaluated in two species of the genus Thlaspi, the Cd hyperaccumulator alpine pennycress (T. caerulescens) and the nonaccumulator field pennycress (T. arvense). Plant seeds were placed in filter paper imbibed in distilled water with 0 and 10 μM Cd(NO3)2 for germination. Six day old plantlets were then grown on Rorison solution supplemented with the same concentrations of cadmium for 22 days. At the end of the exposure, plants were harvested, and root and shoot lengths, as well as Cd accumulation, were estimated. Cd-exposed plants of T. arvense showed chlorotic expanded leaves, lower growth and Cd accumulated mostly in roots. Cd-exposed T. caerulescens presented no significant differences is leaf morphology or growth, and Cd accumulated preferentially in leaves. Genomic DNA was extracted from roots of both species and microsatellites (SSRs) were then amplified in order to evaluate genetic instability, but no genetic instability was observed. Data suggest that for these species and for the Cd exposure conditions tested, Cd does not induce microsatellite instability. However, the use of other molecular markers is required in order to complement this study.

O cádmio é um contaminante extremamente tóxico para o ambiente e para a população em geral. Apresenta efeitos citotóxicos, carcinogénicos e mutagénicos. A mutagénese reflecte instabilidade genética e pode ser avaliada através de técnicas moleculares (e.g. microssatélites). Neste trabalho os efeitos mutagénicos/genotóxicos do cádmio vão ser avaliados em duas espécies de Thlaspi, uma não acumuladora de cádmio (Thlaspi arvense) e outra hiperacumuladora (Thlaspi caerulescens). As sementes foram colocadas em papel de filtro com 0 e 10 μM Cd(NO3)2 de modo a germinarem. Ao sexto dia as plântulas foram transferidas para o meio de Rorison, onde cresceram durante 22 dias em presença das concentrações de cádmio acima referidas. No fim da exposição colheram-se as plantas, mediu-se raiz e parte aérea e fez-se a avaliação da acumulação de cádmio. As plantas da espécie T. arvense expostas a cádmio apresentaram clorose generalizada e necrose nas folhas expandidas, crescimento diminuído e acumulação de cádmio essencialmente na raiz. No caso das plantas de T. caerulescens expostas a cádmio não se observaram diferenças na morfologia da folha nem no crescimento e a acumulação de cádmio ocorreu preferencialmente nas folhas. Fez-se a extracção do DNA genómico da raiz das duas espécies de modo a avaliar a instabilidade de microssatélites. Nas condições do estudo e nos microssatélites analisados não foi detectada instabilidade genética. Contudo o estudo deve ser complementado recorrendo a outros marcadores moleculares.
