Abstract

A presente dissertação contempla o estudo de reacções de substituição nucleófila nos anéis pentafluorofenilo do 5,10,15-tris(pentafluorofenil)corrol e do 5,10,15-tris(pentafluorofenil)corrolatogálio(III)(piridina) com vista à obtenção de novos fotossensibilizadores para utilização em Terapia Fotodinâmica Antimicrobiana. Foram utilizados nucleófilos aromáticos e alifáticos, tendo sido preparados novos derivados corrólicos com ligações do tipo éter, tio-éter e amina. A análise estrutural dos compostos sintetizados foi realizada com recurso a espectroscopia de Ressonância Magnética Nuclear mono e bi-dimensional (RMN de 1H e 19F, COSY, e NOESY), espectrometria de massa em ESI e MALDI, tendo-se recorrido ainda à espectrofotometria de Ultravioleta-Visível (UV-Visível). Foram estudadas as propriedades fotofísicas dos novos derivados, tendo sido determinados vários parâmetros tais como: o rendimento quântico de fluorescência, rendimento quântico de geração de oxigénio singuleto, tempo de vida de fluorescência e tempo de vida do estado excitado tripleto. Foram ainda realizados estudos biológicos preliminares de fotoinactivação da bactéria bioluminescente Vibrio fischeri com os compostos que se revelaram mais promissores a nível fotofísico.

This dissertation reports the synthesis of new corrole derivatives based on nucleophilic substitution reactions involving the pentafluorophenyl moieties of 5,10,15-tris(pentafluorophenyl)corrole and 5,10,15- tris(pentafluorophenyl)corrolategallium(III)(pyridine) aiming to prepare new photosensitizers to be used in Antimicrobial Photodynamic Therapy. Using a variety of aromatic and aliphatic nucleophiles new corrole derivatives with ether, tioether and amine bonds were synthesized. The structure of the new compounds was determined by several spectroscopical techniques, namely, Nuclear Magnetic Resonance spectroscopy mono- and bi-dimensional (1H and 19F NMR, COSY and NOESY), mass spectrometry (MALDI and ESI) and also UV-Visible spectrophotometry. The photophysical properties of the new synthesized compounds were evaluated and some parameters as fluorescence quantum yield, singlet oxygen quantum yield, fluorescence lifetime and triplet excited state lifetime were determined. Preliminary biological studies were also performed with the most promising compounds in order to evaluate their photoinactivation efficacy in bioluminescent bacteria Vibrio fischeri.
