A presença crescente de fármacos em ecossistemas aquáticos, nomeadamente antibióticos como a ciprofloxacina, tem levantado preocupações ambientais e de saúde pública, devido à sua persistência e potenciais efeitos adversos. Este estudo teve como objetivo investigar os efeitos da ciprofloxacina na metilação global do DNA em Daphnia magna, um organismo modelo que desempenha um papel fundamental na investigação de processos ecológicos e na proteção da saúde ambiental, e explorar possíveis relações entre alterações epigenéticas e alterações fenotípicas, nomeadamente na taxa de alimentação. A exposição de D. magna decorreu durante 48 horas, sendo utilizada uma concentração ecotoxicologicamente relevante (Concentração que provoca imobilização em 20% dos indivíduos expostos durante 48 h - EC₂₀ = 19,57 mg/L) de ciprofloxacina. Após o ensaio, extraiuse DNA e RNA das amostras obtidas. A metilação global do DNA foi quantificada por ELISA, e os efeitos fenotípicos foram avaliados através de ensaios de inibição da taxa de alimentação. Os resultados obtidos indicaram que, nas condições testadas, não houve diferenças estatisticamente significativas entre os grupos controlo e exposto, tanto a nível epigenético, como fenotípico. A ausência de efeitos significativos pode estar relacionada com limitações experimentais, como a curta duração da exposição, a baixa solubilidade da ciprofloxacina no meio ASTM e a utilização de um método global de deteção de metilação, pouco sensível a alterações locus-específicas. Assim, os resultados não excluem a possibilidade de alterações epigenéticas mais leves ou localizadas, algo que será confirmado num futuro próximo após a sequenciação de todo o genoma com técnicas que possibilita a deteção das citosinas metiladas e da sua posição no genoma. Paralelamente, o transcriptoma obtido através de RNAseq será também analisado por forma a esclarecer o papel da metilação na regulação da expressão genética em resposta à exposição a ciprofloxacina. Desta forma, este estudo contribui para a compreensão do impacto dos antibióticos em organismos aquáticos e reforça a importância da utilização de metodologias mais sensíveis e abordagens complementares em trabalhos futuros.
The growing presence of pharmaceuticals in aquatic ecosystems, particularly antibiotics such as ciprofloxacin, has raised environmental and public health concerns due to their persistence and potential adverse effects. This study aimed to investigate the effects of ciprofloxacin on global DNA methylation in Daphnia magna, a model organism that has key role in investigating ecological processes and protecting environmental health, and to explore possible relationships between epigenetic alterations and phenotypic changes, particularly in feeding rate. Exposure of D. magna was carried out for 48 hours, using an ecotoxicologically relevant concentration (kthe concentration estimated to elicit 20% of immobilization of exposed organisms in 48 h - EC20 = 19,57 mg/L) of ciprofloxacin. Following the exposure, DNA and RNA were extracted from the samples. Global DNA methylation was quantified by ELISA, and phenotypic effects were assessed using a post-exposure feeding inhibition assay. The results obtained indicated that, under the tested conditions, there were no significant differences between the control and exposed groups, either epigenetically or phenotypically. The absence of significant effects may be related to experimental limitations, such as the short duration of the exposure, the low solubility of ciprofloxacin in ASTM medium and the use of a global methylation detection method, which is not sensitive to locus-specific changes. Thus, the results do not exclude the possibility of subtle or localized epigenetic changes, which is to be confirmed in the near future with the analysis of whole genome methylation sequencing data applied to the DNA samples. The whole transcriptome will also be inspected over RNAseq data in the near future to clarify the role of methylation in regulating gene expression responses to ciprofloxacin stress. The present study thus contributes to the understanding of the impact of antibiotics on aquatic organisms and reinforces the importance of more sensitive methodologies and complementary approaches in future epigenetic investigations.
