Neste estudo, os compostos do café instantâneo solúveis em água fria foram fracionados por ultrafiltração e os extratos obtidos foram caracterizados quanto à composição em polissacarídeos, proteínas, ácidos clorogénicos e melanoidinas. Os extratos foram ainda avaliados quanto às propriedades antioxidantes, imunoestimuladoras e antidiabéticas. Foram utilizadas duas amostras, uma previamente fracionada em 2011, e outra preparada em 2012. Na amostra processada em 2011, os extratos tinham sido exaustivamente lavados com água para minimizar a presença de compostos de baixo peso molecular em extratos de peso molecular mais elevado. Na amostra processada em 2012, os extratos foram somente lavados duas vezes. Os seis extratos obtidos em cada ano foram caracterizados em 2012 quanto à composição química e atividade biológica. Os polissacarídeos representavam 20-50%, constituídos maioritariamente por galactose, manose e arabinose em ligações 1,3-Gal, 1,6-Gal, 1,3,6-Gal, T-Gal, 1,4-Man, 1,4,6-Man, TMan, T-Ara e 1,5-Ara, diagnósticos da presença de arabinogalactanas e galactomananas. Quanto à composição em aminoácidos, não existiu muita variação entre os diferentes extratos, apresentando todos eles uma maior quantidade de ácido glutâmico/glutamina (7,0- 25,6 μg/mg) e leucina (3,7-9,9 μg/mg). Os ácidos cafeoilquínicos (CQA) (187,7-575,0 mg/100g) foram os ácidos clorogénicos mais abundantes em todos os extratos, sendo o 5-CQA o CQA maioritário, com 83,1-242,4 mg/100g de amostra. Na amostra de 2011 os CGA só foram encontrados no extrato inferior a 1 kDa, havendo quantidades residuais nos extratos >1 kDa e >5 kDa. Na amostra de 2012 os CGA foram abundantes em todos os extratos, aumentando a sua quantidade com a diminuição do peso molecular. A presença de melanoidinas foi avaliada pela quantificação de compostos castanhos pelo coeficiente de extinção Kmix. 405 nm e pela determinação do índice de acastanhamento das melanoidinas (MBI), cujo valor é diretamente proporcional ao Kmix. 405 nm e inversamente proporcional à quantidade de compostos desconhecidos na amostra. Os extratos mais castanhos foram os de elevado peso molecular, >100 kDa (1,97 mL.mg-1.cm-1 e 3,31, respetivamente, para a amostra de 2011 e 1,55 mL.mg-1.cm-1 e 3,37, respetivamente, para a amostra de 2012). A cor castanha dos extratos diminuiu com a diminuição do peso molecular. O menor valor de MBI (0,81 para o extrato 1 kDa da amostra de 2011 apresentou um potencial efeito imunoestimulador pois ativou in vitro os linfócitos B de ratinho. Estes resultados foram confirmados por ensaios de produção de óxido nítrico por parte de macrófagos de ratinhos. Para o extrato superior a 100 kDa da amostra de 2012 verificou-se uma diminuição da atividade da α-glucosidase. O presente estudo demonstra que os compostos do café instantâneo podem possuir efeitos bioativos devido às suas propriedades antioxidantes, imunoestimuladoras e antidiabéticas.
In this study, the instant coffee compounds soluble in cold water were fractionated by ultrafiltration and the extracts obtained were characterized according to their composition in polysaccharides, proteins, chlorogenic acids, and melanoidins. The extracts were also evaluated according to their antioxidant, immunostimulatory, and antidiabetic properties. Two samples were used, one previously fractionated in 2001, and another one prepared in 2012. In the sample processed in 2011, the extracts were exhaustively washed with water to minimize the presence of low molecular weight compounds in extracts of higher molecular weight. In the sample processed in 2012, the extracts were washed only twice. The six extracts obtained in each year were characterized in 2012 in relation to their chemical composition and biological activity. Polysaccharides represent 20-50%, consisting mainly of residues of galactose, mannose and arabinose in 1,3-Gal, 1,6-Gal, 1,3,6-Gal, T-Gal, 1,4-Man, 1,4,6-Man, T-Man, T-Ara and 1,5-Ara glycosidic linkages, suggesting the presence of arabinogalactans and galactomannans. The total amino acids amount of all extracts was similar, showing higher values of glutamic acid/glutamine (7.0-25.6 μg/mg) and leucine (3.7-9.9 μg/mg). The caffeoylquinic acids (CQA) (187.7 to 575.0 mg/100g) were the most abundant chlorogenic acids in all the extracts, where the 5-CQA was the major CQA, with 83.1 to 242.4 mg/100 g of sample. In the sample of 2011 the CGA were only found in the extract lower than 1 kDa, with trace amounts in extracts >1 kDa and >5 kDa. In the sample of 2012 the CGA were abundant in all extracts, increasing their amount with the decrease of the molecular weight. The presence of melanoidins was evaluated by quantification of brown compounds through extinction coefficient Kmix 405 nm and by determination of the "Melanoidin Browning Index" (MBI), which is directly proportional to Kmix 405 nm and inversely proportional to the amount of unknown compounds in the sample. The browner extracts were those with the higher molecular weight >100 kDa (1.97 mL.mg-1.cm-1 and 3.31, respectively for the sample of 2011 and 1.55 mL.mg-1.cm-1 and 3.37, respectively for the sample of 2012). The brown colour of the extracts decreased with the decreasing of the molecular weight. The lowest value of MBI (0.81 for the 1 kDa of the 2011 sample presented a potential immunostimulatory effect since it activated in vitro mice B lymphocytes. Assays with nitric oxide production by mice macrophages confirm these results. A decreasing activity of the α-glucosidase was observed with the extract >100 kDa of sample of 2012. The present study shows that the compounds from instant coffee may have bioactive effects due to their antioxidant, immunostimulatory and antidiabetic properties.
