Suplementos nutricionais - a polémica do trispicolinato de crómio(III)

Abstract

O suplemento nutricional tris-picolinato de crómio(III) tem estado, nos últimos anos, no centro de várias polémicas quanto aos seus efeitos na saúde pública. A suplementação com o tris-picolinato de crómio(III) tem sido ativamente promovida devido à sua presumível habilidade de reforçar o metabolismo da glicose e dos lípidos. Por conseguinte, a popularidade aumentou ainda mais uma vez que é visto como um recurso muito vantajoso para melhorar sintomas associados a doenças cardiovasculares, à obesidade e, principalmente, a Diabetes mellitus tipo II. Tendo em conta a ampla utilização de suplementos nutricionais de crómio onde a evidência científica a favor da eficácia e segurança é limitada e controversa, o presente trabalho teve como objetivo principal determinar os efeitos histopatológicos da suplementação de crómio em níveis crescentes sobre a forma de picolinato de crómio. Foram utilizados ratinhos machos, com 7 semanas de idade e com um peso corporal compreendido entre 30 e 40 g, tendo sido divididos em vários grupos. Em dois dos 4 grupos experimentais o suplemento de [Cr(pic)3] foi administrado por via subcutânea, na dose de 5mg/Kg e de 10mg/Kg diariamente durante 7 dias. Nos outros dois grupos experimentais o suplemento de [Cr(pic)3] foi administrado por via oral, na dose de 25mg/Kg e 50mg/Kg, respetivamente, num período de 14 dias. Após o sacrifício dos animais, foram removidos e pesados o fígado, rim, testículo, epidídimo, baço e timo, fixados em solução de Bouin e preparados para estudos histológicos. Não foram evidenciadas diferenças significativas no peso dos animais e respetivos órgãos. Histologicamente, os efeitos do tris-picolinato de crómio(II) foram mais evidentes nos grupos injetados com 10mg/Kg de [Cr(pic)3] e 50mg/Kg de [Cr(pic)3] por via oral. O suplemento de [Cr(pic)3] provocou alterações histológicas em todos os órgãos em estudo. Dada a evidência de lesões histopatológicas sugere-se o desenvolvimento de estudos futuros, visando o melhor conhecimento dos mecanismos de ação deste suplemento.

Nowadays the nutritional supplement chromium(III) picolinate on public health has been a matter under polemic focus. The use of supplements of chromium(III)picolinate is actively promoted due to their ability to reinforce the metabolism of glucose and lipids. Owing to the ability for alleviate symptoms related to several pathologies such as cardiovascular, obesity and mainly Diabetes Mellitus tipe II this compound has a relevant role. However scientific evidence of its efficacy is limited and controversial. Then, the main goal of the present study was to evaluate the histopathological effects of graded supplement of chromium(III) as chromium(III) picolinate. For this purpose, male mice (7 weeks old, weighing 30 and 40g) were used. Animals were divided in different groups: 2 groups were experimentally submitted to subcutaneous injection of 5 mg/Kg and 10 mg/Kg daily, respectively, during seven days. Other groups were given 25 mg/Kg and 50 mg/kg of the supplement orally for 14 days. After sacrifice, organs such as liver, kidney, testis, epididymes, spleen and thymus were removed, fixed in Bouin´s solution and routinely prepared for histology. No significant differences were noted within animals and organs weights. Histopathological effects were evident in all groups and more pronounced changes were observed within groups administered with 10 mg/Kg of [Cr(pic)3] and 50 mg/Kg de [Cr(pic)3] . For this reason, further investigations should be carried out aiming to elucidate the mechanisms underlying this nutritional supplement.
