Pirosecuenciación del Genoma de una Cepa Andina de Potato Virus S (PVS, Carlavirus) Infectando Solanum phureja (Solanaceae) en Colombia

Abstract

El Potato virus S (PVS) es uno de los virus prevalentes en los cultivos de papa del mundo. En Colombia este virus es de los más incidentes en diferentes variedades de S. tuberosum subsp.andigena y S. phureja. Estudios previos basados en análisis de secuencias del gen de la cápside viral (CP) de aislamientos colombianos de PVS, detectaron la presencia de las dos razas del patógeno (PVSO y PVSA), así como un alto nivel de variación entre aislamientos de PVSA. En esta investigación se secuenció el 98% (8330 nt) del genoma de la cepa RL5 de PVS obtenida a partir de una planta naturalmente infectada de S. phureja var. Criolla Colombia en el municipio de La Unión (Antioquia). Para esto se realizó una pirosecuenciación del ADNc obtenido a partir del ARNm de dicha planta, utilizando el sistema 454 Life Sciences (Roche). El genoma del aislamiento RL5 de PVSA (Número de Accesión JX683388) fue ensamblado a partir de un total de 10899 reads, con una profundidad promedio de 470.8 y resultó filogenéticamente relacionado con la cepa BB-AND de PVSA de Brasil, con la que compartió una identidad genética del 94%. Un análisis de agrupamiento basado en el gen CP, permitió identificar a dicha cepa como perteneciente al clado III de PVSA. La información de este genoma puede ser utilizada para el diseño de cebadores y sondas específicas que apoyen los programas de certificación de tubérculo-semilla y manejo del PVS en Colombia.
