Abstract

Se ha estudiado el efecto antinociceptivo derivado del bloqueo de los receptores de quimiocinas CCR1 y CCR2 en el dolor inflamatorio y neoplásico experimental. El aumento local de la concentración de CCL2 en respuesta a una inflamación aguda o crónica contribuye al desarrollo de la hiperalgesia inflamatoria. Por ello, la hiperalgesia puede ser inhibida tanto a través del antagonismo del receptor CCR2 como de la neutralización de la CCL2. Del mismo modo, el bloqueo de los receptores CCR1 contrarrestó la hiperalgesia y en el caso de la inflamación crónica, la alodinia. Dicha inhibición del dolor parece deberse al bloqueo de la acción ejercida por la CCL3. Los efectos antinociceptivos derivados del bloqueo de CCR1 y CCR2 son independientes de un efecto antiinflamatorio. Además, se describen las respuestas hipernociceptivas derivadas de la inoculación intratibial de células de cáncer de próstata RM1 en ratones inmunocompetentes, siendo un modelo experimental útil para el estudio del dolor neoplásico de origen prostático. El bloqueo de CCR2 evita la hiperalgesia térmica en ratones inoculados con células RM1 y se acompaña de un aumento de la quimiocina CCL2.
