Abstract

1. " Hemos investigado la significación clínica del patrón de expresión de cortactina (CTTN), FAK y podoplanina durante el desarrollo y progresión de los carcinomas de laringe, así como la frecuencia de amplificación génica de CTTN y FAK, para establecer su papel en el proceso de transformación maligna. Se analizó mediante inmunohistoquímica la expresión de cortactina, FAK y podoplanina en muestras parafinadas de 82 pacientes con diagnóstico histopatológico de displasia de laringe, así como en el correspondiente tumor desarrollado durante el seguimiento clínico. Encontramos expresión intensa de cortactina, FAK y podoplanina respectivamente en 41 (50%), 40 (49%) y 38 de las displasias de laringe estudiadas. La amplificación génica de CTTN y FAK fue detectada respectivamente en 10 (12%) y 26 (32%) de las displasias laríngeas. Aquellos pacientes cuyas lesiones premalignas presentaban una expresión intensa de cortactina o FAK mostraban una incidencia de cancer significativamente mayor. Además la expresión de FAK fue predictor independiente del riesgo de desarrollar un carcinoma de laringe y la combinación de cortactina y FAK demostró el mayor valor predictivo positivo. Nuestros hallazgos en este proyecto apoyan el papel de cortactina, FAK y podoplanina en la transformación maligna y proporcionan evidencias originales de la potencial utilidad clínica de la expresión de cortactina y FAK como marcadores de riesgo de cáncer de laringe. La expresión de podoplanina se incrementa en los estadios iniciales de la tumorigénesis laríngea y parece asociarse también con un incremento en el riesgo de cáncer de laringe ".
