Abstract

[ES] Las estatinas son medicamentos que se utilizan para reducir los niveles altos de colesterol, inhibiendo su producción en el hígado y reduciendo los niveles de colesterol asociado a lipoproteínas de baja densidad (LDL-C) de la sangre. Estos medicamentos actúan específicamente inhibiendo la enzima hidroxi-3-metil-glutaril coenzima A reductasa (HMGCR) en el hígado, que se encarga de transformar la HMG-CoA en mevalonato mediante una reducción, un paso crítico en la síntesis de colesterol. En este trabajo se recogió información sobre los potenciales riesgos asociados al uso de estatinas y se analizaron las notificaciones de efectos adversos recogidas por la Agencia Española de Medicamentos y Productos Sanitarios (AEMPS). Para ello, se realizó una búsqueda utilizando diversas fuentes de información y priorizando artículos de los últimos 5 años. Existen muchos efectos adversos asociados a las estatinas que tienen poca representación, de entre ellos, los más comunes y documentados, con diferencia, son la aparición de trastornos musculoesqueléticos y del tejido conjuntivo, que se observan sobre todo en personas de entre 18 y 65 años. A pesar de que hay estudios que intentan describir cómo provocan las estatinas este tipo de trastornos, el mecanismo no es totalmente conocido y hay varias propuestas que se recogen en este trabajo para intentar obtener una visión más global de las posibles causas de estos efectos adversos

[EN] Statins are medications used to reduce high cholesterol levels by inhibiting its production in the liver and lowering levels of low-density lipoprotein cholesterol (LDL-C) in the blood. These medications work specifically by inhibiting the enzyme hydroxy-3-methyl-glutaryl coenzyme A reductase (HMGCR) in the liver, which is responsible for converting HMG-CoA into mevalonate through a reduction, a critical step in cholesterol synthesis. This study gathered information on potential risks associated with the use of statins and analyzed reports of adverse effects collected by the Spanish Agency of Medicines and Medical Products (AEMPS). To achieve this, a search was conducted using various sources of information, prioritizing articles from the past 5 years. There are many adverse effects associated with statins that have low representation, among them, the most common and well-documented are musculoskeletal and connective tissue disorders, primarily observed in individuals aged 18 to 65. Despite efforts to describe how statins cause these types of disorders, the mechanism is not fully understood, and several hypotheses are discussed in this study to gain a more global understanding of the potential causes of these adverse effects
