Abstract

[ES] La pared celular de las plantas juega un papel crucial en su defensa contra el ataque de patógenos. Actúa como barrera física y como barrera química activando los mecanismos de defensa inmune de plantas, como la PTI. Los objetivos del trabajo se basaron en la obtención y caracterización de oligosacáridos a partir de la pared celular de maíz, y en la evaluación de la capacidad inmunoestimulante de los mismos. Para ello, se generaron muestras de pared celular de maíz (AIR) o de fracciones hemicelulósicas (fr. KOH) tratadas enzimáticamente con Driselasa y endoxilanasa GH11. El análisis de la composición de dichas fracciones indicó que estaban compuestas mayoritariamente por oligosacáridos de Arabinosa, Xilosa y Galactosa. Por otra parte, mediante la cuantificación de H2O2, por el método del luminol-peroxidasa, se intentó determinar si alguno de los compuestos obtenidos actuaba como DAMP e iniciaba la respuesta defensiva. Sin embargo, los resultados obtenidos no mostraron respuesta de ningún tipo. Es muy posible que se deba a que los oligosacáridos obtenidos de las digestiones enzimáticas eran de tal tamaño que les impedía actuar como DAMPs. Como propuesta de futuro sugeriría repetir el procedimiento experimental con otro tipo de enzimas para intentar caracterizar las moléculas que sí estimulan la PTI ya que, mediante la aplicación externa de moléculas DAMP se podría mejorar la resistencia de los cultivos

[EN] The plant cell wall plays a crucial role in its defence against patogen attacks. It acts as a physical and chemical barrier, activating the immune defence mechanism of the plant, such as the PTI. The present research aimed to obtain and characterize oligosaccharides from maize cell wall and to evaluate their immunostimulant capacity. To achieve so, several maize cell wall (AIR) or hemicellulosic fractions (fr. KOH) samples were generated and enzymatically treated using Driselase and endoxylanase GH11. The analysis of the composition of these fractions indicated that they were mainly composed of oligosaccharides of Arabinose, Xylose, and Galactose. Moreover, quantifying H2O2 through the luminol-peraxodase method aimed at determining whether any of the obtained components acted as DAMP and initiated a defensive response. However, the outcome did not show any response. The oligosaccharides obtained from the enzymatic digestion were of such size that they could not act as DAMPs. The same experimental procedure could be repeated using a different kind of enzyme to try and characterize the molecules that indeed estimulante the PTI since, through the extrernal application of DAMP molecules, the resistance of crops could be increased
