Ritmos circadianos y el cáncer: potencial de la cronoterapia

Abstract

Los ritmos circadianos conforman un mecanismo biológico de coordinación entre multitud de procesos fisiológicos y metabólicos del organismo con respecto a los sucesos del ambiente que lo rodean para adaptarlo de manera eficiente. Si bien su concepto lleva años establecido, su vínculo con la salud y la enfermedad se ha vislumbrado hace pocas décadas. El presente Trabajo de Fin de Grado presenta como hipótesis la existencia de una correlación entre la desregulación circadiana y la progresión tumoral, razón del potencial de la cronoterapia. Para apoyarla, y mediante una extensa revisión bibliográfica, se recoge el funcionamiento a nivel molecular del ritmo circadiano como base para contrastarlo con el desarrollo tumoral, y así establecer cuáles y cómo los diferentes genes del entramado circadiano están involucrados en los procesos celulares que conforman los principales hallmarks del proceso cancerígeno. Se discuten las posibles relaciones causa-efecto entre ambos apoyado también en varios ejemplos. Asimismo, y tomando las nociones de los ritmos circadianos y la cronobiología, se define y se propone el concepto de cronoterapia como método en segunda línea de optimización de la quimioterapia y radioterapia tradicionales contra el cáncer, y para ello se recoge su nacimiento como enfoque alternativo y se exponen varios casos que apoyan su fundamento, así como varios métodos mediante los cuales incrementar su efectividad para, por último, discutir su viabilidad clínica presente y futura.
