Proteómica del endometrio humano

Abstract

La presente tesis doctoral se centra en el estudio del endometrio, el cual desarrolla temporalmente un estado en el que se permite la adhesión del blastocisto después de la fecundación, para posibilitar la implantación. Además este tejido permite que se produzca la posterior invasión controlada del trofoblasto que dará lugar a la placentación, que resulta imprescindible para el desarrollo fetal. Todos esos cambios, tanto a nivel morfológico como molecular, están regulados a nivel proteico y por ello, caracterizar y conocer el proteoma podría ser utilizado para mejorar el entendimiento de este proceso. Esta tesis persigue obtener el conjunto de proteínas diferencialmente expresadas del endometrio humano comparando la fase pre-receptiva con la fase receptiva. Tras ello, se estudian aquellas proteínas que resultan claves en este proceso biológico. Entre ellas, estatmina 1 destacó llamativamente respecto al resto y se expresaba fundamentalmente a nivel del estroma endometrial y por tanto, podría estar regulando un proceso clave como la decidualización. El siguiente objetivo concreto fue caracterizar el patrón de proteínas expresadas tanto intracelularmente como extracelularmente durante dicho proceso. Finalmente, otra proteína llamada anexina A2, destacó respecto al resto por su aumento significativo y consistente durante la receptividad endometrial y por tanto, se llevo a cabo un estudio molecular para averiguar la implicación de esta proteína sobre la implantación embrionaria.
