Abstract

Töö eesmärk on analüüsida Phytophthora liikide levikut puudel ning välja selgitada võimalik Phytophthora levimine taimlatiikides ja jõgedes ning testida Phytophthora liikide DNA praimereid. Välitööde käigus koguti üle Eesti halli lepa, sanglepa, arukase ja paju liikidelt puiduproove, mida testiti Phytophthora spp. kiirtestiga ning puidu- ja leheproovid isoleeriti puhaskultuuri ja eraldati DNA. Taimlatiikidest ja jõgedest Phytophthora määramiseks kasutati püünistena rododendroni lehti. DNA analüüsi käigus Phytophthora liigispetsiifilise praimeritega andsid positiivsed tulemused kolm A. incana puiduproovi 71st. Liikide määramisel tuvastati Põlvamaalt, Kõnnu külast kahest asukohast kogutud puiduproovidest P. alni või P. cambivora. Järvamaalt, Paide vallast kogutud halli lepa proovist määrati perekond Phytophthora spp. (100%) esinemine. Taimlatiikidest ja jõgedest kogutud proovidest andsid positiivse tulemuse kaks liigispetsiifilise praimeriga analüüsitud proovi 5st. Tartu puukooli tiigist kogutud proovist määrati P. gallica (100%) ja Emajõest P. gonapodyideas (99%). Phytophthora praimerite testi tulemusena leiti, et edaspidiseks analüüsideks on soovituslik kasutada liigispetsifiilise praimerit FMPhy-8b; FMPhy-10b.

The purpose of the thesis is to analyze Phytophthora spp. species on trees and also to investigate the reach of the infection in different plant nurseries and rivers in Estonia, also test the species-specific DNA primers. During the fieldwork wood samples were collected from grey alder, silver birch, willow and black alder. The wood samples were analyzed with Phytophthora spp. test, wood and leaves samples were isolated to pure culture and DNA was extracted. To identify Phytophthora spp. in ponds and rivers rhododendron leaves were used. Three grey alder samples provided positive results with species-specific primer (FMPhy- 8b; FMPhy-10b). Two samples were collected from Põlva county, Kõnnu village and identified as P. alni or P. cambivora and one sample from Järva county it showed Phytophthora spp. (100%). Two samples collected from plant nursery ponds and one sample from rivers indicated positive results with species-specific primer. P. gallica (100%) was determined from Tartu plant nursery water pond and P. gonapodyideas (99%) from Emajõgi. Phytophthora primers’ test showed that species-specific primer FMPhy-8b; FMPhy-10b is most suitable to use.
