Abstract

Los neutrófilos juegan un papel crucial en la defensa frente a candidiasis sistémica, una enfermedad asociada con elevadas tasas de mortalidad en pacientes inmunodeprimidos, si bien los mecanismos que el sistema immune emplea para disparar las actividades fungicidas de los neutrófilos aún no se encuentran bien definidos. En este trabajo hemos explorado el papel de los monocitos inflamatorios Ly6Chi en la activación de neutrófilos mediada por células natural killer durante las primeras fases de la respuesta immune innata a la infección por C. albicans, utilizando un modelo múrido de candidiasis sistémica. Nuestros datos sostienen que la defensa eficaz frente a la infección por Candida requirió una respuesta coordinada entre el riñón y el bazo, dependiente de la producción de IL-15 por los monocitos Ly6Chi de bazo, a través de un mecanismo mediado por interferon de tipo I, la cual permitió una eficiente activación y liberación de GM-CSF por las células natural killer de bazo, necesaria para disparar el potencial fungicida de los neutrófilos de riñón. De esta forma, nuestros datos revelan una función hasta ahora desconocida de la IL-15 en la defensa frente a candidiasis sistémica y permite abrir nuevos frentes para el desarrollo de terapias frente a esta enfermedad

Neutrophils play a crucial role in defense against systemic candidiasis, a disease associated with a high mortality rate in patients receiving immunosuppressive therapy, although the early immune mechanisms that boost the candidacidal activity of neutrophils remain to be defined in-depth. In this work, we have explored the role of inflammatory Ly6Chi monocytes in natural killer cell-mediated neutrophil activation during the innate immune response against C. albicans, using a murine model of systemic candidiasis. Our data support that efficient anti- Candida immunity required a collaborative response between the spleen and kidney, relying on type I interferon-dependent IL-15 production by spleen inflammatory Ly6Chi monocytes, driving an efficient activation and GM-CSF release by spleen natural killer cells, that was needed to boost the Candida killing potential of kidney neutrophils. Our data thus unveil an unknown function of IL-15 as a critical mediator in defense against systemic candidiasis, and hold promise for the design of novel IL-15-based antifungal immunotherapies.
