Generación de tejidos artificiales funcionalizados para aplicaciones de ingeniería tisular craneofacial y dental

Abstract

Los avances recientes en ingeniería de tejidos ofrecen alternativas clínicas innovadoras en odontología y medicina regenerativa. La ingeniería tisular combina células humanas con biomateriales compatibles para inducir la regeneración de tejidos. Una de las limitaciones de la ingeniería tisular es el tiempo requerido para fabricar los tejidos artificiales, por lo que reducir dicho tiempo de biofabricación contribuiría significativamente a la mejora del tratamiento. Asimismo, la funcionalización de los biomateriales utilizados en ingeniería tisular mejoraría las propiedades funcionales del tejido artificial y, por tanto, su utilidad clínica. En esta Tesis Doctoral, hemos probado un método de biofabricación alternativo mediante la inclusión directa de bioexplantes de tejido de mucosa oral humana dentro del biomaterial para la generación de tejidos orales y maxilofaciales humanos para su uso en ingeniería tisular. Para ello, se desarrollaron y analizaron biomateriales acelulares de fibrina-agarosa, tejidos artificiales de fibrina-agarosa no funcionalizados que contenían células estromales, y nuevos sustitutos de tejido de fibrina-agarosa funcionalizados con bioexplantes tisulares, analizándose cada tipo de tejido tras 1, 2 y 3 semanas de desarrollo en cultivo. Tras ello, se analizaron los componentes de la matriz extracelular para determinar su nivel de biomimeticidad respecto a los controles de la mucosa oral humana nativa aplicando métodos histológicos, histoquímicos e inmunohistoquímicos. Los resultados mostraron que la funcionalización contribuyó a acelerar y mejorar el método de biofabricación y a obtener un tejido artificial con características biomiméticas respecto al tejido nativo en términos de componentes de la matriz extracelular.

Recent advances in tissue engineering offer innovative clinical alternatives in dentistry and regenerative medicine. Tissue engineering combines human cells with compatible biomaterials to induce tissue regeneration. Shortening the fabrication time of biomaterials used in tissue engineering will contribute to treatment improvement, and biomaterial functionalization can be exploited to enhance scaffold properties. In this work, we have tested an alternative biofabrication method by directly including human oral mucosa tissue explants within the biomaterial for the generation of human bioengineered mouth and dental tissues for use in tissue engineering. To achieve this, acellular fibrin–agarose scaffolds (AFAS), non-functionalized fibrin-agarose oral mucosa stroma substitutes (n-FAOM), and novel functionalized fibrin-agarose oral mucosa stroma substitutes (F-FAOM) were developed and analyzed after 1, 2, and 3 weeks of in vitro development to determine extracellular matrix components as compared to native oral mucosa controls by using histochemistry and immunohistochemistry. Results demonstrate that functionalization speeds up the biofabrication method and contributes to improve the biomimetic characteristics of the scaffold in terms of extracellular matrix components and reduce the time required for in vitro tissue development.
