Genética molecular y evolutiva de los cromosomas B de Prospero autumnale

Abstract

Prospero autumnale (Asparagaceae) es una planta herbácea bulbosa perenne distribuida por toda la región circunmediterránea. Constituye un complejo de especies crípticas con diferentes números cromosómicos y contenido de DNA genómico. Además de esta variabilidad genómica canónica, se han descrito cromosomas B en tres de sus cuatro citotipos diploides. Estos cromosomas son supernumerarios, prescindibles y no se recombinan con los cromosomas estándar (A). Por lo tanto, siguen su propia vía evolutiva, mostrando los mecanismos de impulso en la línea germinal de los individuos que los contienen. En la Cuesta de la Palma (Granada, España), Ruiz-Rejón et al. encontraron en los años 80 del siglo pasado una población de P. autumnale que albergaba cromosomas B que afectaban la expresión de una enzima esterasa. Sin embargo, aún quedaban por conocer muchos aspectos de este sistema de cromosomas B. Hemos realizado una serie de análisis en esta población para abordar algunos temas fundamentales de la investigación del cromosoma B (por ejemplo, evolución y transmisión de frecuencia). Aprovechando el desarrollo de nuevas tecnologías de secuenciación de DNA, también hemos analizado el contenido de DNA de estos cromosomas B.

Prospero autumnale (Asparagaceae) is a perennial bulbous herbaceous plant distributed throughout the circum-Mediterranean region. It constitutes a complex of cryptic species with different chromosomal numbers and genomic DNA content. In addition to this canonical genomic variability, B chromosomes have been described in three of their four diploid cytotypes. These chromosomes are supernumerary, dispensable, and do not recombine with the standard (A) chromosomes. Therefore, they follow their own evolutionary pathway, showing drive mechanisms in the germline of the individuals containing them. At Cuesta de la Palma (Granada, Spain), Ruiz-Rejón et al. found in the 80s of the last century a natural population of P. autumnale harbouring B chromosomes that affected the expression of an esterase enzyme. However, many aspects of this B chromosome system yet remained to be known. We have performed a series of analyses in this population to address some fundamental topics of B chromosome research (e.g., frequency evolution and transmission). Taking advantage of the development of new DNA sequencing technologies, we have also analyzed the DNA content of these B chromosomes.
