Toxicity of many xenobiotics is mediated by oxygen free radical production. These xenobiotics are reduced in oneelectron reactions to give rise to chemical especies which react with molecular oxygen (O2) producing superoxide anion radicals (O2.-) and regenerating the original molecule. In this way, a redox cycling is formed involving disproportionate consumption of O2 and redox equivalents, mainly NADPH. Radicals produced during this redox cycling, and their derivative structures, would be responsible for the cellular damage caused by such xenobiotics.
