Abstract

La criptosporidiosis es una zoonosis, de gran importancia en la población infantil y en la inmunodepresión, frente a la que no existe una terapia especifica. partiendo del cultivo de c.parvum sobre celulas mdck, se ha llevado a cabo la evaluación de la actividad de distintos farmacos in vitro frente a cryptosporidium.los ooquistes de cryptosporidium se obtienen inicialmente de heces de terneras con parasitacion espontánea y se someten a un proceso de concentración y purificación según metodología propia, y a digestión para liberar los esporozoitos. la interacción parasito-célula se lleva a cabo en proporción 1:1 durante 12 horas. la metodología empleada conduce a un porcentaje de parasitación de un 8% siendo las distintas formas de desarrollo del parasito visualizables a microscopia óptica, mediante técnicas especiales de tincion. la acción de los distintos fármacos sobre el sistema parasito-célula se ensayaron a distintos tiempos, tanto en la fase de interacción, (acción preventiva), como en cultivos establecidos, (hasta 72 horas), (acción curativa-inhibición de la merogonia continuada). así se realizo un estudio que se centra en: la determinación de la actividad in vitro de antiprotozoarios conocidos por su demostrada acción frente a otros coccidios. la determinación de la actividad in vitro de antiprotozoarios de nueva síntesis, compuestos acridinicos y sus derivados. selección de los compuestos mas efectivos y estudio de citotoxicidad de estos sobre células mdck y de la relación estructura-actividad
