Actividad nitrato reductasa y monóxido de carbono deshidrogenasa en bradyrhizobium japonicum

Abstract

En esta tesis doctoral se ha llevado a cabo la secuenciación y caracterización del gen afectado en el mutante grf3, que es deficiente en la expresión del enzima nitrato reductasa y de membrana. La secuenciación de la zona afectada en la mutación ha dado lugar a la localización de genes coxsmorf4orf5 que codificarían genes relacionados con el enzima monóxido de carbono deshidrogenasa en b. japonicum. Se han encontrado también orfs, orf6, 7 y 8, de función desconocida que pudieran estar formando parte de un mismo operon junto con los genes de la utilización de monóxido de carbono. En esa misma región y en sentido de la transcripción contrario a genes que codificarían para el enzima co deshidrogenasa, se han localizado los genes pcabdc que intervienen en la degradación de compuestos aromáticos del tipo del protocaehuato. Se ha llevado cabo en b. japonicum la purificación a homogeneidad electroforetica de un enzima monóxido de carbono deshidrogenasa. Así como la capacidad de dicha bacteria para utilizar el co como única fuente de carbono y energía
