Abstract

En esta tesis se ha estudiado el catabolismo de la prolina en P.putida KT2440. El estudio incluye el aislamiento y secuenciación de los genes putA y putP responsables del catabolismo de la prolina en este organismo, así como el aislamiento y caracterización de mutantes deficientes en la lutilizaciónd e este compuesto. Tras la secuenciación del operón se caracterizó la expresión de los genes implicados en el catabolismo de la prolina en distintas condiciones y fondos genéticos (IHF, sigma 38 y PtsN). Los promotores del operón se utilizaron para la construcción del elemento regulador de un sistema de contención biológica dependiente de prolina., basado en la fusión de los promotores PputA y PputP al gen lacl. Tras la inserciónestable del elemento regulador a través de un minitransposón en una cepa de P.putida KT2440, se indtrodujo el elemento matador basado en la fusión de unpromotor derivado del promotor Plac y el gen suicida (genE) del fago X174, tras la obtención de la cepa suicida se comprobó el correcto funcionamiento del sistema de contención condicionada por la presencia de prolina en el medio
