Biofluorescência no meio marinho: estrutura e propriedades de uma Pioverdina

Abstract

Os complexos bactéria-esponja são actualmente uma área de investigação importante tanto do ponto de vista da Biologia Marinha como da Química, uma vez que permitiram a identificação de inúmeros produtos naturais com potenciais aplicações tecnológicas e farmacêuticas. Neste trabalho identificou-se uma estirpe de Pseudomonas putida associada ao complexo marinho Cliona viridis, colhida na reserva natural da Berlenga. A estirpe bacteriana designada por NB3L foi isolada e cultivada em laboratório, o seu rDNA 16S extraído e sequenciado. A estirpe NB3L apresenta fluorescência típica de várias espécies de Pseudomonas. A molécula responsável por esta fluorescência foi purificada, tendo sido desenvolvido um protocolo de purificação simples, rápido e eficaz, diferente dos descritos para compostos desta família. Tendo por base resultados de espectroscopia vibracional no infravermelho, espectrometria de massa e espectroscopia de ressonância magnética nuclear, a molécula foi identificada como uma pioverdina. Esta família de sideróforos pode também estar envolvida em comunicação intercelular. Os resultados obtidos sugerem que se trata duma nova pioverdina com algumas propriedades distintas das descritas anteriormente, como sejam uma elevada hidrofilicidade, precipitação a pH > 10.5, componentes de decaimento de intensidade de fluorescência com tempo de vida mais longo, e uma maior fotoestabilidade em solução. O coeficiente de absorção molar, a posição do espectro de fluorescência e o rendimento quântico de fluorescência são dependentes do pH e de iões metálicos. Verificou-se uma interacção específica entre esta pioverdina e os iões metálicos Fe3+, Al3+ e Cu2+. Um estudo mais aprofundado da estirpe NB3L e da sua pioverdina, incluindo as propriedades de fluorescência e sua função, poderão contribuir para esclarecer as interacções bactéria-esponja nos ecossistemas.

Sponge-associated bacteria have been an important topic of research in the past few years. These studies have contributed to the fundamental knowledge of Marine Biology and Chemistry, and allowed the identification of inumerous new natural products with potential technology and health related applications. In the present work, we report on the identification of a bacteria of the genus Pseudomonas associated with the sponge Cliona viridis collected on the Portuguese coast. The bacterial strain, currently designated NB3, was isolated and cultured in laboratory, and the 16S rDNA was extracted and sequenced. NB3 produces fluorescence typical of several Pseudomonas species. The molecule responsible for the fluorescence was purified, by a new protocol easier, faster and more efficient than other methods described for these compounds. Taking into account results from vibracional infrared spectroscopy, mass spectrometry and nuclear magnetic ressonance spectroscopy methods, the molecule was identified as a pyoverdin. This is a family of siderophores that may also be involved in intercellular communication. This molecule has apparently both chemical and photophysical properties that are unique among pyoverdins. Some of these features include a very high hydrophilicity, precipitation at pH > 10.5, absence of a pronounced photobleaching under continuous illumination, and the high photostability in solution. The molar absorption coefficient, position of the fluorescence spectra, the quantum yield and fluorescence lifetime are pH and metal ion dependent. A specific interaction between this pyoverdin and the metal ions Fe3+, Al3+ and Cu2+ was found. Further studies on the NB3L strain and its pyoverdin, including fluorescence properties and function, will hopefully shed light into novel roles of sponge-bacteria association in ecosystems.
