Mediadores e inflamação alérgica

Abstract

O estudo dos mediadores e da sua determinação como marcadores de inflamação tem adquirido novas aplicações com a introdução de recentes tecnologias. Como marcadores gerais de inflamação são em geral utilizadas as moléculas de adesão, para os mastocitos a triptase, para os eosinófilos a proteína catiónica dos eosinofilos — ECP — para leucócitos circulantes incluindo os basofilos o doseamento in vitro após contacto com o antigénio dos leucotrienos (CAST), e como marcador especifico de activação dos basofilos a citometria de fluxo (FAST), utilizando uma dupla marcação por antiIgE (basófilos e anti CD63) marcador de activação. Nesta revisão serão apresentados resultados obtidos nos últimos anos pelo nosso grupo de trabalho no estudo da triptase no lavado nasal, o ECP no lavado nasal, as moleculas de adesão solúveis, a libertação de leucotrienos por método imunoenzimático — ELISA,e a activação dos basófilos por citometria de fluxo (FAST). Estes métodos podem ser utilizados tanto para diagnóstico etiológico como para demonstração de reactividade celular ou para controlo de terapêutica tanto farmacológica como imunoterapia específica.
