Abstract

Os anticorpos, mais propriamente os anticorpos monoclonais, pertencem a uma classe relativamente nova, sendo considerados uma estratégia bem sucedida no tratamento de diversos tumores sólidos e de neoplasias do foro hematológico. O seu desenvolvimento constitui um dos maiores avanços dos últimos anos e o sucesso na terapêutica envolve um processo complexo de conhecimentos científicos e avaliações pré-clínicas bem como uma compreensão profunda da biologia do cancro e das propriedades dos anticorpos. Em geral os anticorpos são administrados em combinação com quimioterapia e/ou radioterapia, embora nalgumas circunstâncias possam ser usados em monoterapia. Baseiam-se na capacidade de reconhecer antigénios específicos de tumores e induzir uma resposta imune contra as células malignas. Além disso, os anticorpos monoclonais podem ser modificados de forma a actuarem como portadores de radioisótopos ou agentes citotóxicos – anticorpos conjugados – ampliando o seu espectro de aplicação terapêutica. Anticorpos biespecíficos, isto é, anticorpos que reconhecem dois antigénios diferentes, também constituem uma estratégia eficaz. Os inúmeros ensaios clínicos que envolvem anticorpos também são de extrema relevância e poderão constituir, num futuro próximo, alternativas viáveis à radioterapia e quimioterapia, que estão associadas a reacções adversas e toxicidade significativas.

Antibodies, more known as monoclonal antibodies, belong to a relatively new class and are considered a successful strategy in the treatment of various solid tumours and haematological malignancies. Their development is one of the greatest advances in recent years and the success in therapy involves a complex process of scientific knowledge and pre-clinical reviews, as well as a deep understanding of cancer biology and properties of antibodies. In general, antibodies are administered in combination with chemotherapy and/or radiotherapy, although in some circumstances they can be used alone. They´re based on the ability to recognize tumour-specific antigens and induce an immune response against malignant cells. Furthermore, monoclonal antibodies can be modified to act as carriers of radioisotopes or cytotoxic agents – conjugated antibodies – broadening its spectrum of therapeutic application. Bispecific antibodies are antibodies that recognize two different antigens, are also an effective strategy. The numerous clinical trials involving antibodies are also extremely important and may be, in the near future, viable alternatives to radiotherapy and chemotherapy, which are associated with significant adverse reactions and toxicity.
