Efeitos da Ribavirina (Virazole) a ultrastrutura nuclear em células CV1

Abstract

A Ribavirina (Virazole) tem sido estudada como antiviral, não se sabendo ainda bem se o seu mecanismo de acção é especificamente antiviral ou se a inibição da replicação viral é o resultado do seu efeito tóxico na célula hospedeira. Os seus efeitos no metabolismo celular estão pouco esclarecidos. Não conhecemos nenhum trabalho sobre os efeitos da Ribavirina na ultrastrutura celular. Em resultados preliminares do tratamento de células CV1 com várias concentrações de Ribavirina observámos alterações marcadas na ultrastrutura nucleolar, tais como: segregação, aumento de volume e extrusão dos centros fibrilhares e fragmentação do nucléolo com aparecimento de nódulos fibrilhares no espaço intercromatínico. Ë de frisar que estas alterações são evidentes mesmo a concentrações baixas da Ribavirina (50 jjg/ml). Observou-se mesmo um efeito de certo modo independente da dose do antimetabolito, pois as alterações observadas a tempos de exposição ás 3 concentrações da Ribavirina não apresentaram diferenças ultrastruturais significativas. Efeitos morfológicos semelhantes a estes foram vistos em células tratadas com outros antimetabolitos e em muitos casos são devidos a uma inibição da síntese dos percursores do ARN nucleolar. O mecanismo mais provável da acção da Ribavirina é a inibição do enzima IMP dehidrogenase, que bloqueia a biosíntese dos nucleótidos da guanina. Tal microtrauma bioquímico pode estar na base das alterações ultrastuturais do nucléolo evidenciadas pelas imagens.

La Ribavirine (Virazole) a été etudiée en tant qu’agent antivirale, mais on ne sais pas encore bien si le mecanisme d’action est specifiquement antivirale ou si l’inibition de la replication virale est le résultat de son effect toxique sur la ceilule infectée. Les effects sur le metabolisme cellulaire sont peu connus. Nous ne connaissons aucun travail sur les effects de la Ribavirine sur l’ultrastructure cellulalre. Dans des études préliminaires sur l’effect de différentes concentrations de Ribavirine sur des cultures des cellules CV1 nous avons observé des modifications sur l’ultrastructure nucléolaire: ségregation, augmentation du volume et extrusion des centres fibrillaires dans l’espace intercromatinique. Ces modifications sont évidentes à partir de 50 jig/ml de Ribavirine et sont independentes de la concentration du anti-métabolite car les différences observeés aux temps d’exposition dans les trois concentrations de la Ribavirine n’avaient pas differences ultrastructurales significatives. Ont etés observés des effects morfologiques sínifiaires SUT des celiules traitées avec d’autres anti-métabolites et dans beaucoup de cas ils sont conséquence d’une inibition de la synthèse des percurseurs de l’ARN nucléolaire. Le mecanisme plus évident de l’action de la Ribawirlne est l’imbition de l’enzime IMP dehidrogenase, quiempèche ia biosyntesse des nucléoUdes de la guanine. Les différences ultrastructurales nucléolaires observées peuvent être du à cet effect.

Ribavirin (Virazole) lias been studied like antiviral, but it is not yeat clear whether the compound is specfflcally antiviral in its mode of action or whetlier It inhibits virus replication as a result of its effect(s) on the host cell. The effects on cellular metabolism are not well known we dont’t know any work about the effects of Ribavirin on the ceflular ultrastructure. In preliminar results, treatment of CV1 culture celis with Ribavirin ia varying doses produced marked alterations in nucleolar ultrastructural organization. A series of changes occur including segregation, an increase of the volume e extrusion of the fibrillar centers and fragmentation of nucleolus with appearence of fibriliar nodules in the intereromatin space. Low concentrations of Ribavirin (50 g/ml) lead the sarne alterations than higher concentr’ations, to the sarne period of time. Similar ultrastructural lesions are seen lii treated celis with other antimetabolites and lii several cases, they are resultei of the inibition of the synthesis of nucleolar percursor RNA. The possible mecanism of Ribavirin is the inibition of the enzime inosine-5’-monophosphate (IMP) dehydrogenase that blocks tlie biosynthesis of nucleotides. This effect can be responsable fot the images seen.
