Abstract

A ß-talassemia é uma doença caracterizada pela diminuição da síntese das cadeias de ß-globina. Nas suas formas mais graves pode causar falência multiorgânica, associando-se a uma morbilidade e mortalidade elevadas. No presente trabalho procedeu-se a uma revisão da bibliografia publicada até ao momento acerca desta doença, sendo as informações retiradas de livros, websites e bases de dados digitais. Pretendeu-se explorar a etiologia e a sintomatologia inerentes à ß-talassemia, identificar as terapêuticas mais utilizadas e, também, fármacos inovadores. A etiologia da doença assenta em alterações moleculares cuja heterogeneidade é evidente. Reconhece-se um elevado espetro de manifestações clínicas, desde portadores assintomáticos até doentes com sintomas severos. As terapêuticas mais utilizadas são a transfusão sanguínea e a quelação do ferro. Atualmente, encontram-se em estudo terapêuticas de reversão do silenciamento dos genes fetais, de controlo do stress oxidativo e de manipulação genética. Estes estudos poderão vir a ser importantes para contornar as desvantagens inerentes às terapêuticas mais utilizadas, tanto a nível clínico como económico. Os programas de prevenção são também relevantes no controlo da doença.
