Abstract

During vertebrate embryonic development the presomitic mesoderm (PSM) is gradually divided into segments, the somites. In the chick embryo one pair of somites forms every 90 minutes. It is generally thought that the tight temporal and spatial control of somitogenesis is regulated by the cyclic expression of genes of the Notch and Wnt signaling pathways, and the response of their protein products to retinoic acid and FGF/Wnt gradients. The cellular reorganization involved in somitogenesis requires adhesion to the extracellular matrix particularly to fibronectin (FN). In addition to regulating the cellular dynamics underlying the morphological segmentation of the PSM, preliminary data indicated that it may also be involved in its molecular segmentation. The objective of this thesis project was to address this issue. Through the application of inhibitors of FN fibrillogenesis and Notch signaling in cultures of embryo explants and isolated PSMs and the subsequent assessment of mRNA expression patterns, it was revealed that the FN matrix is required for segmentation both at the morphological level and at the level of the expression of delta1 and meso1. Furthermore, the effect of inhibiting FN fibrillogenesis produced a phenotype that was very similar to the one obtained upon inhibiting Notch signaling. It was found that Notch signaling activates meso1 expression which in turn regulates segmentation and that absence of Notch signaling and a FN matrix cause a similar disturbance in the expression of delta1 and meso1. Finally it was observed that molecular segmentation appears to be not only the result of intrinsic PSM signaling, but is also influenced by ectoderm, as evidenced by the requirement for exogenous FN for molecular segmentation in the rostral PSM and for delta1 expression in the caudal PSM.
