Abstract

A Nanotecnologia é uma ciência com uma abordagem revolucionária que desenvolve produtos e processos tecnológicos em nanoescala. Oferece a oportunidade destas partículas serem transportadas seletivamente para locais alvo, melhorando a biodisponibilidade dos fármacos e reduzindo a sua toxicidade e efeitos colaterais. A administração cutânea de fármacos tem vindo a tornar-se numa estratégia importante para superar aspetos indesejados relacionados com as características farmacocinéticas e farmacodinâmicas dos fármacos. A pele é uma via de administração que apresenta várias vantagens mas que se depara consecutivamente com a mesma dificuldade, a impermeabilidade parcial da pele, devido às várias camadas da pele que formam uma barreira à penetração de fármacos, sendo que apenas uma pequena fração da dose atinge o alvo. Desta forma, têm vindo a ser desenvolvidos métodos para uma melhor penetração na pele. Esta presente Dissertação de Mestrado tem como principal objetivo relatar sumariamente as diferentes estratégias usadas para promover a absorção cutânea de fármacos, identificando as principais barreiras, vantagens e os produtos de sucesso alcançados até ao momento presente.

Nanotechnology is a science with a revolutionary approach that develops technological products and processes at the nanoscale. It offers the opportunity for these particles to be transported selectively to target sites, improving the bioavailability of the drugs and reducing their toxicity and side effects. Cutaneous administration of drugs has become an important strategy for overcoming unwanted aspects related to the pharmacokinetic and pharmacodynamic characteristics of the drugs. Skin is a route of administration which has several advantages but which faces the same difficulty, limited permeability of the skin, due to the various layers of the skin that form a barrier to the penetration of drugs, and only a small fraction of the dose reaches the target. In this way, methods for better skin penetration have been developed. This master thesis has as main objective to briefly report the different strategies used to promote cutaneous absorption of drugs, identifying the main barriers, advantages and the most representative products of success until this date.
