Abstract

Neste trabalho foi estudada a Paropsia brazzeana Bail, uma planta medicinal angolana recolhida na Huila. O material vegetal foi macerado e extraído a frio com metanol. De seguida procedeuse ao processo de fracionamento do extrato bruto em fração de hexano/acetato de etilo 95:5, hexano/acetato de etilo 8:2, hexano/acetato de etilo 1:1, acetato de etilo, acetato de etilo/metanol 9:1, acetato de etilo/metanol 7:3, acetato de etilo/metanol 1:1, metanol e metanol/ácido clorídrico. Das frações obtidas com as misturas de vários solventes com polaridades crescentes, foram isolados quatros compostos. O geraniol foi isolado a partir da fração hexano/acetato de etilo 95:5, o ß-sitosterol e o ácido octanóico foram isolados a partir da fração hexano/acetato de etilo 1:1 e o ácido 14- nor-7,8-seco-10-oxocadinano-1(6),3-dienóico, que isolado a partir da fração acetato etilo, foi isolado pela primeira vez no género Paropsia e foi considerado um novo produto natural. Nas restantes frações não foi isolado nenhum composto. As frações hexano/acetato de etilo 95:5, hexano/acetato de etilo 8:2, hexano/acetato de etilo 1:1 e acetato de etilo foram utilizadas para testes de citotoxicidade nas células MCF-7 e NHDF. Para efetuar os ensaios de citotoxicidade fez-se o ensaio para determinar a viabilidade celular (MTT) e o ensaio de doseamento de proteínas (BCA). Os resultados dos ensaios de viabilidade celular e de doseamento de proteínas feitos nas MCF-7 mostraram que todas as frações testadas aparentemente são tóxicas para essas células. Os resultados dos ensaios feitos nas NHDF são semelhantes aos resultados obtidos nas MCF-7, tanto os ensaios da viabilidade celular como os resultados do doseamento de proteínas apontaram para uma possível toxicidade das frações para com as células.

This master thesis reports the study of Paropsia brazzeana Bail, an Angolan medicinal plant collected in Huila. The plant material was macerated and extracted with cold methanol giving the crude extract. Then proceeded to the crude extract fractionation process obtaining the following fractions: hexane/ethyl acetate 95:5, 8:2, 1:1 hexane/ethyl acetate fraction; ethyl acetate fraction; ethyl acetate/methanol fraction 9:1, 7:3; 1:1, 7:3 ethyl acetate/methanol fraction; ethyl acetate/methanol 1:1; methanol and methanol/hydrochloric acid. Different fractions eluted with various mixtures of solvents with increasing polarity afforded four compounds. Geraniol was isolated from the fraction of hexane/ethyl acetate 95:5. ß-sitosterol and octanoic acid were isolated from the fraction hexane/ethyl acetate 1:1. 14-Nor-7,8-seco-1(6), oxocadinanedienoic acid was first isolated in Paropsia genus and was considered a new natural product, this compound was isolated from the ethyl acetate fraction. In the remaining fractions wasn’t isolated any compound. The fractions hexane/ethyl acetate 95:5, hexane/ethyl acetate 8:2, hexane/ethyl acetate 1:1 and ethyl acetate were used in cytotoxicity assays in MCF-7 and NHDF cells. To perform the cytotoxicity assays the assay was made the cell viability test (MTT) and the protein assay (BCA). The results of cell viability assays and protein determination made in MCF-7 showed that all extracts tested are apparently toxic to these cells. The results of the tests made in NHDF are similar to the results obtained in MCF-7, both the tests of cell viability such as protein assay results pointed to a possible toxicity of the extract towards the cells.
