Abstract

O cancro atualmente é considerado uma das principais causas de morte em todo o mundo, sendo que a sua incidência continua a aumentar, apesar de todos os avanços científicos ao longo dos anos. Atualmente métodos como a cirurgia, radioterapia e a terapia sistémica são usados para tratar e controlar a doença. Contudo, a investigação na descoberta de novos fármacos é fundamental para que o tratamento desta patologia se torne cada vez mais eficaz. Neste contexto, não só vários esteroides têm mostrado ação antiproliferativa e pró-apoptótica em diversos estudos, mas também várias poliaminas são capazes de encaminhar o composto para o interior das células cancerígenas e favorecem a sua interação com o DNA. Desta forma, o objetivo deste estudo foi sintetizar novos compostos com ação anticancerígena com possível interação com o G-quadruplex envolvendo a combinação de poliaminas com estruturas esteroides. Desta forma, foram preparadas e caracterizadas várias poliaminas esteroides e a sua ação anti-proliferativa, bem como a dos seus intermediários sintéticos foram avaliadas em células cancerígenas da mama (MCF-7, T47D), da próstata (LNCaP), do glioblastoma (U87) e em células normais (NHDF) pelo ensaio MTT, de modo a se concluir a cerca do seu potencial interesse anticancerígeno. Neste contexto, e no sentido de explorar o seu mecanismo de acção, também foram realizados ensaios de fluorescência e de citometria de fluxo. Assim, da síntese química resultaram cinco compostos cujas ações antiproliferativas foram posteriormente estudadas. Destes, mediante o ensaio MTT, o composto c8 foi o que originou os melhores resultados, evidenciando não só uma acção marcada contra a linha de cancro da próstata, com um IC50= 1,368, bem como uma selectividade relevante em comparação com as células normais (IC50= 31,75). Mediante estudos de citometria de fluxo, realizada com este composto em células MCF-7, verificou-se não existirem valores significativos de morte celular. Quanto ao ensaio de fluorescence resonance energy transfer, os compostos C1, C3, C4 e C7 não evidenciaram interação com o G-quadruplex.

Cancer today is considered to be one of the main causes of death anywhere in the world. It’s a disease that, in spite of all the scientific advances along the years, continues to spread. Nowadays methods like surgery, radiotherapy and systemic therapy are used to treat and control this illness. However, the development of new drugs remains crucial to the treatment of all forms of cancer. Various steroids have shown antiproliferative and pro-apoptotic properties in numerous studies. Many polyamines have the capability to route compounds to the interior of the cancer cells and to favour their interaction with the DNA. Having this in mind, the aim of this project is to synthesize new anti-cancer compounds that can possibly interact with G-quadruplex and that involve the combination of polyamines with steroid structures. Thus, several steroidal polyamines were synthesized and their antiproliferative properties, as well as those of their intermediate synthetics, were evaluated in cancerous breast cells (MCF-7, T47D), cancerous prostate cells (LNCaP), glioblastoma cells (U87) and in non-tumoral cells (NHDF). This was done recurring to the MTT assay in order to study the potential anti-cancer properties of these compounds. In this context and in order to explore their action mechanism, fluorescence and flow cytometry studies were also conducted. From the chemical synthesis resulted five compounds that were posteriorly studied. Regarding the MTT study, the compound with which the best results were observed was C8 that has shown, not only meaningful action against the prostate cancer line with a IC50= 1,368, but also relevant selectivity when compared with the non-tumoral cells whose IC50= 31,75. The flow cytometry study conducted on this compound for the MCF-7 cells revealed low cell death values. Concerning the fluorescence resonance energy transfer study, the compounds C1, C3, C4 and C7 did not show interaction with the G-quadruplex.
